Migrasome Purification Kits

For production and purification of cultured-cell and tissue-derived migrasomes.

MigraPure™︎ Purification - Cultured-Cell Migrasome Purification Kit

PRC003S

A kit family for further purification of crude cultured-cell migrasome preparations.

Product specifications

Use this page for product selection and experiment planning. Consult the PRC003 series manual for the full procedure and limitations.

This product is intended for further purification of previously obtained crude migrasome preparations from cultured cells. It does not include upstream cell culture, migrasome production, or crude isolation from cultured cells.

Migratory adherent cell lines or primary cells are recommended, including cells whose adhesion and migration increase after overexpression of Tspan4 or other genes that promote migrasome formation. Migrasome production depends on the cells and must be evaluated experimentally.

PRODUCT FEATURES

Product advantages

Purification of crude preparations

Further separates cultured-cell migrasome crude preparations to obtain purified migrasomes of consistent quality.

Conventional density-gradient separation

Uses differences in component density to separate migrasomes from membrane fragments, organelles, and other impurities, reducing protein contamination.

Adaptable to different cellular sources

Sampling individual gradient fractions helps identify the optimal enrichment layer for migrasomes from different cells.

Broad downstream applications

Purified samples can be used for migrasome counting, protein and RNA analysis, and cell function assays.

FAQ

Frequently asked questions

Does PRC003 include migrasome production and crude isolation from cultured cells?

No. The product starts from an existing crude cultured-cell migrasome preparation and is used for subsequent refinement and purification. Upstream production and crude isolation require a separate validated workflow.

Which cell sources are most suitable for evaluation?

Migratory adherent cell lines or primary cells are recommended, including cells overexpressing Tspan4 or other genes that promote migrasome formation. Suspension cells are not recommended, although extracellular matrix coating or gene editing may be tested to promote adhesion. Optimize density, production time and detachment conditions, and consider in situ WGA staining before routine production, as recommended in the manual.

Which downstream studies may use the purified sample?

Purified samples can be used for migrasome counting, protein and RNA extraction and analysis, and cell function assays. Storage and sample-purity requirements differ among downstream assays. For details, consult the product manual or the Migrasome Therapeutics technical team.

How should purified migrasomes be stored?

For studies of nucleic acid or protein contents, the pellet may be stored at -80°C after high-speed centrifugation and supernatant removal. For morphology, number, surface properties, or activity, storage at 4°C is recommended; storage at 4°C for up to one week has relatively little effect on morphology and number. Freezing at -20°C or -80°C causes aggregation and some rupture. Preliminary experiments are recommended if freezing is necessary.

How should the gradient fractions be selected?

Collecting layers 3–5 in 500 μL fractions is recommended. Enriched layers may differ among cell sources. When isolating migrasomes from a cell line for the first time, retain and analyze samples from each layer to identify the target fractions.
